Forest berries, vegetables, fruit and red vine in big quantities contain extracts of anthocyanins, which have a possibility to destroy effectively cancer cells without healthy cells damage. Anthocyanin-containing extracts we have got can underlie innovational, untoxic preparations as alternatives of chemotherapy on the different stages of cancer deceases. In our research we used anthocyanin extracts SAG-7/bc and SAG-7/bb that were not mattered before. Extracts were obtained from currant and blackberry. As research model we used rats of Wistar population with mass of 180–210 gr. and adenocarcinoma and as research materials there were the affected tissue parts of decapitated animals. Preparations and control solution NaCl of 0,85 % were injected to laboratory animals with probe peroral (per os) in volume 2,0 ml on the basis of 3,0 gr. of extract per 100,0 ml of water (dose 0,3 gr./kg). During already first three days of inoculated cancerous tumour evolution research in vivo we observed that to the 90 % of its colony were ruined. Thus, there was true anticancer
effect of SAG-7/bc and SAG-7/bb extracts. Further increasing of SAG-7/bc and SAG-7/bb extracts concentration led to all cancer cells death. Cell lines of other types of cancerous tumours reacted to anthocyanin extracts in the same way. During studying the anthocyanin extracts influence upon cancer cells we established that given matter from the one hand activates effect functions of immunocompetenced cells and from the other hand causes the beginning of programmed processes of cancer cells death - apoptosis. At the same time, anthocyanin extracts SAG-7/bc and SAG-7/bb influence upon normal tissue cells didn’t cause such effect. On the contrary, the anthocyanin extracts SAG-7/bc and SAG-7/bb defended healthy cells from the negative effect of free radicals. In general, the stable tendency to the abortion of cancer tumour was established.
The analysis of antocyanine glycosides and aglycones summary content have been carried out. As the maximum of long-wavelength band in absorption spectra of the sum of black currant anthocyanines was observed at 538 nm, we have used a reference substance – cyanine having the band maximum at 535 nm. The reference substances in the analysis of flavonoid sum (excluding anthocyanes, which are also flavonoids), carotenoids and chlorophylls were quercetin, β-carotene and chlorophyll A, correspondingly. The analysis of antioxidant activity has been also carried out. The analyses of the sums of anthocyanes, flavonoids, carotenoids and chlorophylls were made using the spectrophotometry; the estimation of antioxidant activity was carried out using chemiluminescence decay kinetics. The spectrophotometer Hitachi U3210 was used for spectral analyses, and the spectrofluorimeter Hitachi F4010 was applied for chemiluminescence detection.
The concentration of anthocyanines was determined according to a method, described in [Roshal A.D., Tsynovy V.I., Orlov V.D., Patent N. 1744647, Bull. invent. – 1992. – No 24]. Antioxidant activity of extracts was estimated by chemiluminescence decays of phenyl ester of acridiniumcarboxyle acid according to [Krzymiński K., Roshal A.D., Synchykova O.P., Sandomirsky B.P., Patent P–381661, Poland, prior. 01.02.2007]. The data listed in the table show that obtained extract contains approximately 93 % of anthocyanines. Moreover, the extract contains 4.85 mg/g of other flavonoids. The presence of substances mentioned above explains the antioxidant and P-vitamin activity of the extract. The estimation of antioxidant activity was carried out using rate constants of chemiluminescence decays. The value of partial chemiluminescence decay constant is 15.5 1/g/s. The results of the chemical analysis of the anthocyanine extract are listed in table 1.
The antioxidant activity of anthocyanine extract from black currant (1) in comparison with the same parameter of «ether» and lipophil coffee extracts (2) and (3), quercetin (4), 10% aqueous solution of ascorbic acid (5), ascorutin (6) are presented on figure 2. The figure shows that antioxidant activity of anthocyanine extract is close to that of other powerful antioxidants. Summarizing results mentioned above, we can conclude that anthocyanine extract from black currant has a high concentration of biologically active flavonoids, which explains its substantial antioxidant activity and other important therapeutic effects as strengthening vascular system, cancerostatic effect, etc.
